TI HIV antibody responses determined by the ELISA sandwich capture assay
using recombinant glutathione-S-transferase-fused antigens.

AB OBJECTIVE: To develop an ELISA system with a low background level which
enables to quantify the HIV-specific antibodies using E. coli-derived
recombinant antigens. METHODS: HIV-1 gag p17, p24, and pol integrase
were expressed as the glutathione-S-transferase (GST) fusion proteins.
The reombinant antigens partially purified by a glutachione affinity
chromatography were immobilized on microtiter plates which had been
coated with the monoclonal antibody to GST (1G05). Antibody levels in a
total of 147 sera from 36 HIV-1-infected patients and 45 healthy donors
were determined by the improved sandwich capture ELISA. RESULTS: The
ELISA system significantly distinguished between HIV-positive group and
healthy group (p < 0.0001). There were significant correlations among
the kinetics of anti-p17, p24, integrase, and CD4+ cell ratios in
certain HIV-1-infected patients during the course of infection (p <
0.01). In some cases, however, no significant correlations were observed
among all antibodies. CD4+ cell ratios are not always correlate with
kinetics of the antibody responses during our observation period.
CONCLUSION: Our ELISA system provides a fast and relatively cheap
diagnostic tool to determine the HIV-specific antibody titers in human
sera.
